Quality of oak acorns is often affected by the presence of fungal organisms, especially during storage. The aim of the study was to isolate fungi on the agar medium and to identify pure mycelia cultures with molecular methods to determine is there a difference in the species present on control and treated acorns. Pure mycelia cultures obtained from acorn groups treated at different temperatures, different time periods and stored at different temperatures were grouped into morphotypes based on the morphological features observed with the naked eye and microscope (Olympus BX41). For each of the established morphotypes, one representative isolate was chosen for the identification of species by molecular methods, including DNA isolation from mycelium, amplification of target DNA region in PCR reaction, PCR product electrophoresis on agarose gel and sequencing of the resulting PCR product. Based on the results obtained, conclusions can be drawn on the efficacy of thermotherapy in reducing the number of potential pathogens in acorns. Research has shown that fungi were present on a certain share of the acorns in spite of thermotherapy, confirming their role as an important biotic factor affecting the health and germination of acorns. In laboratory conditions, the influence of thermotherapy temperature and duration on embryo mortality and seed germination according to ISTA rules was investigated. At the 52 °C treatment, the germination rate of the acorns was only 3.33%, that is, only 1 irregular seedling was grown from 30 acorns, so it can be said that this is the upper temperature limit of the embryo mortality. Acorns treated at temperatures of 41 °C and 45 °C for 2.5 and 5.0 h and stored at -1 °C and + 3 °C were sawn in nursery conditions in the spring of 2018. The article presents the results of nursery seed germination and morphological properties of 1 + 0 bare roots seedlings. All of the seedlings have been measured or evaluated for 16 morphological features which describe them, in order to determine are there statistically significant differences between treatments.
