Comparison of standard clotting tests in dogs between QuickVet and ACL 7000 analyzers

Standard clotting tests for evaluation the functionality of the blood coagulation cascade are prothrombin time (PT) and activated partial thromboplastin time (aPTT). PT is used to assess the extrinsic and common pathway and aPTT to assess the intrinsic and common pathway. The aim of this study was to compare results of standard clotting tests in dogs measured immediately and individually by QuickVet analyzer with those measured in stored samples in one turn by ACL 7000 analyzer. The QuickVet analyzer is a mechanical clot detection system based on one-time-use test cartridges containing micro channels and test-specific reagents. However the ACL 7000 analyzer utilizes photo-optical turbidimetric method to measure standard clotting tests. Material for comparison was residual blood from 120 dogs with various medical conditions. Dogs included in the study were admitted at the Clinic for Internal Diseases, Faculty of Veterinary Medicine, University of Zagreb. Blood was drawn only to asses the health status of the dogs. The first measurement was made by the QuickVet analyzer in whole citrated blood. After centrifugation plasma was separated and frozen on -80˚C. Samples were collected for 5 months untill the second measurement on the ACL 7000 analyzer in Special Hospital for Medical Rehabilitation of Stubičke Toplice. The results were divided into two groups of different sizes and different duration of collection. Agreements between measurements were analysed using Bland Altman method and Passing and Bablok regression analysis. PT showed significant constant and significant proportional difference in each group. APTV showed significant proportional difference in each group. However aPTV showed no significant constant difference between two methods in each group. In conclussion methods can not be used interchangeably. Taking into account the analyzing a large number of stored samples at once we recommend measurement by QuickVet analyzer despite the high price of the test.
