C type Lectin Receptor Expressions in Mature Synovial Tissue of Patients with Osteoarthritis
Introduction: A family of C-type lectin receptors, including NKG2A, NKG2C and NK2D are expressed predominantly on the surface of NK cells and a subset of CD8+ T - cells. These receptors are efficient pattern-recognition receptors (PRRs) that interact with ligands via carbohydrate structures, leading to modulation of T and NK cell differentiation. Proteins that contain C-type lectin domains have a diverse range of functions, including cril-eell adhesion, Immune response to antigens and apoptosls. NKG2A inhibits, while NKG2C and NKG2D stimulate cytotoxic activity of NK cells. We investigated the presence arid tissue distribution of C-type lectin family receptors NKG2A, MKG2C, NKG2D in synovial tissue of patients with osteoarthritis (OA) in respect to tissue distribution Of T and NK cells. Material and Methods: Synovial tissue samples (no. 5) were obtained during total knee replacement surgery and embedded in paraffin cubes. Tissue sections were prepared as 5-6 urn thick cuts and labeled by single immunofluorescence using anti- NKG-2A (clone 131411, R&D: Systems, 0, 2 p.g/sample, dilution 1:100), anti-NKG-2C(clone 134522, R&D Systems, 2 jig/sample, dilution 1:100), anti-NKG-2D (clone 1D11, BD Pharmingen, 10 pg/sample, dilution 1:100), anti-CD8 (clone RPA-T8, BD Pharmingen, 20 ul/ slide) and' anti- CD 56 (clone: N901, IgGl ; Beckman Coulter), images were taken using an Olympus BX51 fluorescent microscope and Cell F program version 3.1 (Olymjjus, Tokyo, Japan). Microphotographs were randomly chosen Results: NKG2A and NKG2C were mostly scattered in sublining synovial tissue, resembling the CD8+ and CP56+ tissue distribution. NKG2D was present on the surface of synovial shoots and in the accumulations of lymphocytes. The distribution of this receptor resembles the most the distribution of synovial CD56+ cells. Conclusion: The presence of C-type lectin receptors NKG2A, NKG2C and NKG2D in synovial tissue of patients wjith'mature OA suggests that they might be involved in immunopathogenesis of the disease.
