Cellular responses to proteome-wide isoleucine mistranslation
We investigated an effect of two different types of substitutions: canonical (Ile-to-Val) and noncanonical (Ile-to-Nva) on the E. coli proteostasis machinery. We investigated cellular responses to mistranslation by in vivo experiments and with methods of quantitative proteomics to identify changes in the proteome and DnaK clients isolated in pull-down assay. Proteome analysis revealed significant upregulation of several chaperone systems but not proteases. However, mistranslation did not change the identity of DnaK clients and no noticeable difference between cellular responses to canonical and noncanonical mistranslation was observed.
