Phagocytic activity in F1 hybrid mice injected with parental spleen cells

Parental CBA spleen cells were intravenously injected into untreated adult (C57BL X CBA-T6T6) F1 hybrids in doses of 5, 10 or 20 x197 viable cells. Weight and cellularity of the spleen, origin of diving cells within the spleen, and phagocytic activity were determined in recipient nice during thevcourse of 60 days after parental cells injection. A extensive proliferation of both donor and host cells in the spleen resulting in splenomegaly of parental cells. The incidence of donor cells, the wxtent of atrophy of the white pulp and necrosesof the spkenic tissue weredepebdent on the number of injected parental cells. In F1 hybrids injected with 5 or 10 x 10000000 CBA cells two peaks of phagocytic activity were found. The first one was concomitant with intensive proliferation of cells in the spleen, and it was ascribed primarily to intrasplenic phagocytosis. The second incrase appeared when the spleen was empty and was, therefore, assumed to be due to extrasplenic phagocytosis. The injection od 20 x 10000000 cells produced a severe damage in the recipients spleen. In this case a single increase of the phagocytic activity was observed, whith was ascribed to extrasplenic phagocytosis. It is concluded that splenomegaly im F1 recipients of CBA cells is primarily caused by proliferation of the recipient's cells. Forthermore, it is posible that among these histiocytes prevail and that they are responsible for the increase in phagocytic activity of the F2 hosts.
