Biological activity of Helichrysum italicum and Salvia officinalis essential oils

Aromatic plants growing in Dalmatia coastline Helichrysum italicum (Roth) G. Don subb. italicum and Salvia officinalis L. are traditionally broadly used and their essential oils might be potential source of novel biologically active constituents. Essential oils have been used for skincare purposes. By our present knowledge, there is no scientific research of the biological effect on skin cells of the H. italicum and S. officinalis essential oils. In our research we have tested 7 samples of S. officinalis and 11 samples of H. italicum essential oils on human fibroblasts HF77FA. Cell proliferation assay was performed at density of 8 000 cells/well. The cells were treated with concentrations of essential oils from 10 - 1000 *10 -6 g/L in triplicate for 48 hours. Following the treatment with essential oils, cells were incubated with MTS and measured spectrophotometrically at 492nm. Cytotoxicity LC 5O values ranged from 200 to more than 1000 g/L for H. italicum, while S. officinalis showed cytotoxic range of 442-587 g/L. The results in general showed there is no significant cytotoxic activity on fibroblast cells from both of plant species essential oils. This supports the traditional use of the essential oil of H. italicum and S. officinalis. It has to be point out that in our research we have managed to show the cytotoxic activity of some oils of H. italicum that could be reached with concentrations of oil at 200 g/L. It is important to say that each of the samples was sampled in different location and due to that fact and microclimate conditions had different chemical composition. Different chemical composition is usually connected with different biological activity. Our work proved correlation of chemical composition of the essential oils and their cytotoxicity where the main constituents showed significant correlation to toxicity. The research was supported by Student Council of University of Split and Croatian Science Foundation project IP-2014- 09-6897.
