Differential effect of raw soybean diet on hepatic and whole body glucose homeostasis in rat
The liver plays a key role in glucose homeostasis, lipid, and energy metabolism. Dysregulation is a major cause for the development of the metabolic syndrome and type 2 diabetes. Studies have shown a beneficial effect of soy intake on glycemic control and weight. The aim was to evaluate the effect of soybean diet on body weight, some biochemical parameters in blood, hepatic regulation of glucose production and storage and insulin sensitivity in rats. In vivo studies were performed on male Wistar rats given ad libitum raw soybean (SO) or standard diet (CD), for 3 weeks. Body weight was evaluated. Glucose tolerance test was performed and 14C-glucose incorporation to glycogen was measured in vitro in cultured hepatocytes isolated by collagenase perfusion technique and cultured for 24 h in M199 serum- free medium.Despite a favorable effect soybean diet on glucose homeostasis and weight, in vitro experiments in hepatocytes demonstrated impaired glucose and glycogen metabolism and severe hepatic insulin resistance as reflected by suppression of glucose production and stimulation of glycogen synthesis
