Changes within lymphatic tissue of recipient mice reacting to foreign skin grafts of differing incompatibility

Changes within lymphatic tissue of recipient mice after skin grafting and their dependence on the level of histoincompatibility between the donor and the recipient of the graft were investigated. Female mice of the C57BL strain were grafted with the skin of: hamsters (xenogeneic combination), CBA mice (allogeneic combination), C57BL male mice (allogeneic 'weak' combination), or C57BL female mice (syngeneic combination). During three weeks after transplantation 3 to 6 recipients were killed at regular intervals and in each combination. Regional and contralateral lymph nodes and the spleen were taken, weighed and processed for histology. In the syngeneic combination there were neither signs of graft rejection nor significant changes in the lymphatic tissue. Recipients of xenogeneic and allogeneic grafts displayed enlargement of regional lymph nodes and the spleen in the course of the second and the third week after transplantation. The enlargement was best expressed in mice receiving hamster's skin (xenogeneic combination), while it was somewhat less pronounced in recipients of allogeneic grafts. In the paracortical zone of the regional lymph nodes of recipients of allogeneic skin there were up to 10 times more immunoblasts than in the syngeneic combination. The number of immunoblasts in the allogeneic 'weak' combination was only slightly higher than in syngeneic controls. The maximum number of immunoblasts was found 7 days after transplantation. Changes in the lymphatic tissue of mice transplanted with skin grafts were directly proportional to the level of histoincompatibility in the donor and the recipient of the graft.
