Assessment of criteria for reactivity to H-Y antigen in skin graft rejection experiments
Evaluation of male (H-Y) skin isograft susvival in females of some inbred stains of mice were reexamined science discrepancies of the results found in the literature may be due to different criteria of reejection. Two main criteria of reejection were comsidered: the "firts sign" and the "ultimate sign". Extensive testing of the fate of the first-set and secomd-set H-Y isografts was performed in four inbred stains: C57Bk/GoZgr (H-2b), BALB/cBkl (H-2d), C3Hf/BuZgr (H-2k) and CBA/HZgr (H-2k). Full-thickness male tail skin isografts were transplanted to the torax of females. After analysis of gross appearance and histology of the graftsm the most appropriate criterion for rejection and points was formulated. The H-Y isografts was considered rejected if 90% of its surface necrotized within three weeks after transplantaton, or if, during 70 days, the hair crop wasreduced to an area of about 10% of the initial surfacw. With this criterion C57BL females were responders, BALB and C3H females were intermediate responders, and CBA female were nonresponders to the HY antigen. Some others criteria couse different classification but they produce oversimplifications, ognoring significant percentage of actually immunoreactive recipients.
